Form
Liquid or Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer
If the delivery form is liquid, the default storage buffer is Tris/PBS-based buffer, 5%-50% glycerol.
Note: If you have any special requirement for the glycerol content, please remark when you place the order.
If the delivery form is lyophilized powder, the buffer before lyophilization is Tris/PBS-based buffer, 6% Trehalose, pH 8.0.
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Description

The production of this Recombinant Human ERAS protein started with the ERAS gene synthesis. And then using recombinant DNA technology, the ERAS gene was inserted into an expression vector so that we could get the recombinant express plasmid of ERAS. Transform the plasmid into the cells of E.coli, culture the cells and we could get the desired Recombinant Human ERAS protein. But the work was not completed, protein purification and a strict QC system were performed in the last step. The purity is 90%+ determined by SDS-PAGE.

ERAS (also named HRAS2 or HRASP) is a gene providing an instruction of making a protein named GTPase ERas (ERas) in human. The protein encoded by this gene is also known as Embryonic stem cell-expressed Ras and belongs to Ras family found in murine embryonic stem (ES) cells in 2003. Increasing studies has been reported that ERAS may provide resistance to chemotherapy in different human tumorigenic cells through activation of the PI3K/AKT/mTOR pathway, promoting both DNA replication and transcription.

Target Background

Function
Ras proteins bind GDP/GTP and possess intrinsic GTPase activity. Plays an important role in the tumor-like growth properties of embryonic stem cells.
Gene References into Functions
  1. N-terminal extension of E-RAS is important for E-RAS signaling activity. PMID: 25940089
  2. Expression of ERAS in human melanoma cell lines conferred resistance to PLX4720 and induced hyperphosphorylation of AKT. PMID: 25624498
  3. After knocking down the ERas gene by siRNA, we observed that there was a significant decrease in proliferation, metastasis as well as clonality in gastric carcinoma. PMID: 23612786
  4. Data indicate that expression of ERAS, LHX1, and CCRK is increased in aggressive subgroups of medulloblastomas. PMID: 22875024
  5. ERas may be a potential biomarker for gastric cancer, but advanced studies are wanted. PMID: 21151392
  6. These data suggest that ERas is activated in a significant population of gastric cancer, where it may play a crucial role in gastric cancer cell survival and metastases to liver via down-regulation of E-cadherin. PMID: 20566745
  7. ERas/PI3K pathway may provide resistance to chemotherapy and promote transforming activity in neuroblastoma. PMID: 20811723
  8. transforming oncogene ERas is important in the tumour-like growth properties of embryonic stem cells PMID: 12774123
  9. Studies on ERas gene expression. PMID: 16081664
  10. rasH2 may have a role in susceptibility to chemically-induced tongue and esophageal neoplasms PMID: 18174262
  11. High ERas is associated with the tumorigenic process of gastric carcinomas. PMID: 19528480
  12. Loss of methylation in the promoter of ERas might be one of mechanisms responsible for the re-expression of an embryonic oncogene in gastric cancer. PMID: 19787253
