The Human Haptoglobin (Hpt/HP) ELISA kit is specifically designed and validated to quantitatively detect the levels of HP in the serum, plasma, or tissue homogenates. It is not intended for diagnostic use. This assay kit was designed and optimized for cardiovascular research use in humans. The kit has undergone rigorous quality control in multiple parameters, including sensitivity, specificity, precision, linearity, recovery, and inter-batch difference. Refer to the product instructions for more details.

This assay is based on the Sandwich-ELISA mechanism (the HP in the sample is sandwiched between the pre-coated HP antibody and HRP-labeled HP antibody) and enzyme-substrate chromogenic reaction. The color intensity is positively related to the amount of HP bound in the initial step. The levels of HP in the samples can be determined by referring to the O.D. (optical density) of the samples to the standard curve.

HP is an immunoglobulin (Ig)-like plasma protein primarily produced by the liver. It is crucial for the clearance of free hemoglobin (Hb) and the neutralization of oxidative damage. HP rapidly binds to Hb to form the HP-Hb complex, which is removed through the reticuloendothelial system and receptor-mediated endocytosis via CD163 on hepatocytes, Kupffer cells, and tissue macrophages. This process also prevents iron loss through the kidneys and protects the kidneys from hemoglobin-causing oxidative damage. A rapid reduction of serum HP levels in this process makes serum HP level become a sensitive marker for the presence of free Hb during intravascular hemolysis. Serum HP levels can also become increased in infection and inflammation.

To assess the linearity of the assay, samples were spiked with high concentrations of human Haptoglobin various matrices and diluted with the Sample Diluent to produce samples with values within the dynamic range of the assay.

These standard curves are provided for demonstration only. A standard curve should be generated for each set of samples assayed.


Materials provided
  • A micro ELISA plate---The 96-well plate has been pre-coated with an anti-human HP antibody.
  • One vial standard (1 x 100 μl) ---Dilute a bottle of the standard at dilution series, read the OD values, and then draw a standard curve.
  • One vial HRP-conjugated HP antibody (100 x concentrate) (120 μl/bottle) ---Bind to the HP in the samples or standards and react with the substrate to make the solution chromogenic.
  • Two vials Sample Diluent (20 ml/bottle) ---Dilute the sample solution.
  • One vial Wash Buffer (25x concentrate) (20ml/bottle) ---Wash away unbound or free substances.
  • TMB Substrate (1 x 10 ml) ---Act as the chromogenic agent. TMB interacts with HRP, eliciting the solution turns blue.
  • One vial Stop Solution (10ml/bottle) ---Stop the color reaction. The solution color immediately turns from blue to yellow.
  • Four Adhesive Strips (For 96 wells) ---Cover the microplate when incubation.
  • An instruction manual

As a result of hemolysis, hemoglobin is found to accumulate in the kidney and is secreted in the urine. Haptoglobin captures, and combines with free plasma hemoglobin to allow hepatic recycling of heme iron and to prevent kidney damage. Haptoglobin also acts as an Antimicrobial; Antioxidant, has antibacterial activity and plays a role in modulating many aspects of the acute phase response. Hemoglobin/haptoglobin complexes are rapidely cleared by the macrophage CD163 scavenger receptor expressed on the surface of liver Kupfer cells through an endocytic lysosomal degradation pathway.
