CUSABIO's rabbit TNFα ELISA kit is an in vitro enzyme-linked immunosorbent assay for the quantitative determination of TNFα concentrations in serum, plasma, tissue homogenates, and cell lysates. This assay exclusively recognizes rabbit TNFα. The quantitative sandwich ELISA technique of this kit is based on TNFα antibody-TNFα antigen interactions and an HRP colorimetric detection system to detect the levels of TNFα in samples. The intensity of the color is positively proportional to the amount of bound TNFα in the initial step.
TNFα, commonly referred to as TNF, is a pleiotropic cytokine that plays an essential role in the innate and adaptive immune systems. It is also involved in tissue regeneration, and repair, as well as the modulation of embryonic development, the sleep-wake cycle, lymph node follicle, and germinal center formation. TNF signals through TNFR1 and TNFR2, activate NF-κB and MAPK signaling pathways regulating cell proliferation, survival, differentiation, and apoptosis. Due to its pro-inflammatory properties, TNF is implicated in inflammatory diseases, such as rheumatoid arthritis, Crohn's disease, and psoriatic arthritis. Increased TNF expression has been found in various cancer types. Especially, in breast cancer, TNF is associated with enhanced tumor cell proliferation, higher malignancy grade, increased occurrence of metastasis, and generally poor prognosis for the patient.

Review: I tried the kit to determine the best working dilution (starting concentration 100mg/mL). In the case of liver the best result was obtained with 5x-10x dilution, and for intestine the best results were using undiluted sample to 2x dilution.

Review: I used CSB-E06998Rb to detect the plasma of the model group before operation, 30min-2 hours during operation, and 24 hours after operation. The R2 of the four-parameter model fitted was 0.99868 (see the picture for details), and the standard curve was in line with the expectation. The TNF-a level of 0-2h was in line with the increasing trend. After literature review, the model needed 6h to detect a large number of increased TNF-a inflammatory factor levels, and the inflammatory factor levels of 6h and above would be measured in the future. The research group brother recommends that there are rabbit species, and the price is affordable, and the test results are accurate.

Review: I used CSB-E06998Rb to measure the plasma of the model group before operation, 30min-2 hours during operation, and 24 hours after operation. The R2 of the standard curve fitted by the four-parameter model was 0.99783 (see the picture for details). The standard curve was in line with the expectation and the OD value was in the range of 0 to 3.6. The level of histone was about 900 before injection, increased to about 1100 2 hours after injection, and returned to the initial level 24 hours after injection. I can\'t give you the specific data, because I want to put it in the article, hope to understand! Review: I used CSB-E06998Rb to measure the plasma of the histone injection model group before operation, 30min-2 hours during operation and 24 hours after operation. The R2 of the standard curve fitted by the four-parameter model was only 0.68, while the R2 of the straight line fitting was 0.9996 (see the picture for details of the data and the fitting curve equation). The standard curve was made in two columns. As expected, the OD values were in the range of 0 to 3.2, and the data obtained were in line with the expected trend, with a preoperative measurement of about 600 to 800pg/ml. It gradually increased to 1400pg/ml during the operation, and decreased to about 1000 pg/ml 24 hours after the operation. The test of the kit was stable. Two repeated holes were made, and the error was small but the color could be displayed, and the individual difference of rabbits was large.

Review: I used CSB-E06998Rb to measure normal plasma, histone stimulation and co-incubation of materials and plasma. The standard curve was fitted by a four-parameter model, and the R2 was 0.99814 (see picture). The standard curve was made into two columns, and the standard curve met the expectation and the OD value was in the range of 0 to 1.6, and the data were in line with the expected trend. The release of inflammatory factors after histone stimulation was about 2 times that of the normal group. The kit was stable in test and all showed color development.
