At present, the mainstream saying that the spike (S) protein of SARS-CoV-2 is a key protein that mediates virus invasion into host cells. SARS-CoV-2 finds host cell receptors and binds to them through S protein, and is endocytosed into cells through clathrin-mediated membrane fusion. Moreover, the relationship and affinity of host cells and virus surface protein ligands determine the infectivity and cytophilic of the virus, and the pathogenicity is related to the host cell type and function it invades. Regarding the targets of SARS-CoV-2 invasion into human body, the latest research supports ACE2 and CD147.
Angiotensin-converting enzyme 2 (ACE2) is currently the most recognized target for SARS-CoV-2 invasion into human. In January 2020, scientists confirmed that ACE2 is a receptor for the SARS-CoV-2 S protein by crystal structure analysis. And the affinity between ACE2 and the RBD (receptor binding domain) of SARS-CoV-2 is 10 to 20 times higher than that of SARS RBD, which is why SARS-CoV-2 is so infectious. However, ACE2 is usually expressed only in heart, kidney, and vascular endothelial cells. In patients with COVID-19, the respiratory tract, digestive tract, liver, and spleen are often damaged. Moreover, viral nucleic acid detected in nasopharyngeal swabs, saliva, urine, feces, and blood, which suggests that SARS-CoV-2 has other receptors in humans.
During the process of severe acute respiratory syndrome (SARS) coronavirus invasion into host cells, CD147 molecule can mediate the SARS-CoV invasion through interaction with CyPA, as well as its similar mechanism in HIV-1 invasion. CD147-antagonist peptide 9 has a high binding rate to HEK293 cells and has an inhibitory effect on SARS-CoV. Because SARS-CoV-2 and SARS-CoV have similar characteristics, Chen Zhinan's research team investigated the possible role of CD147 in SARS-CoV-2 invasion of host cells.
On March 14, 2020, an article entitled "SARS-CoV-2 invades host cells via a novel route: CD147-spike protein" was published on the preprinted BioRxiv. The results of this experiment indicate that blocking CD147 on host cells has an inhibitory effect on neocrown virus, and that CD147 plays an important role in promoting the invasion of host cells by the virus. Although CD147 can be expressed in multiple tissues and organs, such as malignant tumors, central nervous system, basal layer of epidermal cells, outer root sheath cells of sperm, sperm head, blood system, digestive system and urinary system, it can participate in tumorigenesis, tissue Reconstruction, lymphocyte response, spermatogenesis, viral infection, and regulation of nervous system function in early development. Does this mean that SARS-CoV-2 has no other targets when it invades the human body?
In order to verify this, on March 22, 2020, 94 experts including David E. Gordon, et al. published an online article in the preprinted BioRxiv, screening potential therapeutic targets through the interaction between the SARS-CoV-2 protein and human protein. The author of the article is from the University of California, San Francisco (one of the important centers of the New Crown Virus Research in the United States) and other units. The article was entitled “A SARS-CoV-2-Human Protein-Protein Interaction Map Reveals Drug Targets and Potential Drug-Repurposing”.
In fact, the idea of the article is very simple, but the workload is extremely huge. Researchers cloned, labeled, and expressed 26 of the 29 proteins of SARS-CoV-2 in human cells, and identified human proteins that interact with each protein by affinity purification mass spectrometry (AP-MS). A total of 332 highly trusted human proteins (PPIs) interacting with viral proteins were identified, of which 66 could be potential targets. These proteins can be identified by 69 existing FDA-approved drugs, drugs in clinical trials, and preclinical compound targeting. The identification of host-dependent factor-mediated viral infections may provide important molecular targets for the development of a broad and effective antiviral therapy against SARS-CoV-2 and other lethal coronavirus strains [5].
Sequence analysis of the SARS-CoV-2 isolated showed that the 30kb genome encodes up to 14 open reading frames (Orfs). The 5'Orf1a/Orf1ab open reading frame can encode multiple proteins, which are auto-proteolytically processed into 16 non-structural proteins (Nsp1-16) which form the replicase / transcriptase complex (RTC). RTC consists of a variety of enzymes, including including the papain-like protease (Nsp3), the main protease (Nsp5), the Nsp7-Nsp8 primase complex, the primary RNA-dependent RNA polymerase (Nsp12), a helicase/triphosphatase (Nsp13), an exoribonuclease (Nsp14), an endonuclease (Nsp15), and N7- and 2'O-methyltransferases (Nsp10/Nsp16).
At the 3'end of the viral genome, up to 13 Orf were expressed from 9 predicted sub-genomic RNAs. These include four structural proteins: S, E, M, and N, and nine putative cofactors. In terms of genetic composition, the SARS-CoV-2 genome is very similar to SARS-CoV: each has an Orf1ab that encodes 16 predicted Nsps, and each has four typical coronavirus structural proteins. However, they differ in their complement of 3'open reading frames: SARS-CoV-2 possesses an Orf3b and Orf10 with limited detectable protein homology to SARS-CoV, and its Orf8 is intact while SARS-CoV encodes Orf8a and Orf8b [6] [7] [8].
