MICA is a stress-induced ligand for the activating receptor NKG2D, making it a critical mediator of NK cell and cytotoxic T cell surveillance against tumors and infected cells. This recombinant human MICA construct (aa 24–308) carries a C-terminal Fc tag and demonstrates strong binding to mouse NKG2D in functional ELISA with an EC50 no higher than 10 ng/ml, providing a reliable positive control for NKG2D–ligand interaction studies and receptor-blocking assays. Mammalian cell expression preserves native glycosylation patterns essential for proper MICA folding and receptor engagement, supporting use in cell-based NK activation assays, antibody development campaigns targeting the MICA ectodomain, and surface plasmon resonance binding kinetics experiments. Purity exceeding 95% by SDS-PAGE combined with endotoxin levels below 1.0 EU/μg satisfies the criteria typical for functional cell-based assays where endotoxin-driven background activation could otherwise confound NKG2D-dependent readouts.
