This Human COR ELISA Kit was designed for the quantitative measurement of Human COR protein in serum, plasma, cell culture supernates, tissue homogenates. It is a Competitive ELISA kit, its detection range is 0.049 ng/mL-200 ng/mL and the sensitivity is 0.049 ng/mL.

Three samples of known concentration were tested twenty times on one plate to assess.

Three samples of known concentration were tested in twenty assays to assess.    
To assess the linearity of the assay, samples were spiked with high concentrations of human cortisol in various matrices and diluted with the Sample Diluent to produce samples with values within the dynamic range of the assay.

The recovery of human cortisol spiked to levels throughout the range of the assay in various matrices was evaluated. Samples were diluted prior to assay as directed in the Sample Preparation section.

Materials provided
    • A 96-well Assay plate -- The 96-well plate has been pre-coated with anti-human COR antibody.
    • Two vails Lyophilized Standard -- Dilute one of the standards at dilution series, read the OD values, and then draw a standard curve.
    • Anti-human COR Antibody (100 x concentrate) 1 x 60 μl-- Act as the detection antibody.
    • Antibody Diluent 1 x 10 ml -- Dilute the COR antibody solution.
    • HRP-conjugate (100 x concentrate) 1 x 120 μl-- Bind to the COR antibody, and HRP catalyzes the substrate A and B to elicit a chromogenic reaction.
    • HRP-conjugate Diluent 1 x 20 ml -- Dilute the HRP-conjugate solution.
    • Sample Diluent 2 x 20 ml -- Dilute the standard and sample solutions. Wash Buffer (25 x concentrate) 1 x 20 ml -- Wash away unbound or free substances.
    • TMB Substrate 1 x 10 ml -- HRP catalyzes the TMB, eliciting a chromogenic reaction.
    • Stop Solution 1 x 10 ml -- Stop the color reaction. The solution color immediately turns from blue to yellow.

I would like to know if corticosterone also affects to OD value.

This kit is specific to Cortisol.It has no correlation with corticosterone and the corticosterone does not affect OD value.
